微胶囊和微乳技术对天然抑菌物质抑菌行为影响研究

摘  要:天然抑菌成分主要为一些植物的精油和植物代谢产物。天然抑菌物质拥有安全性高的优点,但是其分散性差,热不稳定,易挥发等缺点,一直是妨碍其实际应用的重大障碍。微乳液和微胶囊技术则可以极大的改善这些缺点。本文以薰衣草精油和水杨酸两种天然抑菌成分作为出发物,制备微乳液以及微胶囊,研究了薰衣草精油,薰衣草精油微乳液,水杨酸和水杨酸微胶囊对金黄色葡萄球菌的抑制率,动态杀菌曲线以及动态细胞膜破坏曲线。抑菌实验的结果表明,薰衣草精油,薰衣草精油微乳液,水杨酸和水杨酸微胶囊均有较好的抑菌能力。其中水杨酸微胶囊的抑菌能力最为突出,在水杨酸浓度为0.125%对金黄色葡萄球菌的抑制率达到了99.9999%。而在400ul/l和800ul/l的浓度下,薰衣草精油以及微乳液也可以抑制1到2个数量级的金黄色葡萄球菌。动态杀菌实验的结果显示,当金黄色葡萄球菌暴露在水杨酸中1min既可以降低4个数量级,而暴露时间达到30min可以降低5个数量级。通过动态细胞膜破坏实验可以得出结论,水杨酸微胶囊拥有缓释效果。上述结果表明,通过微乳液以及微胶囊技术改善天然抑菌物质应用性差的方法可能是天然抑菌物质在实用化领域的一种有效途径。

Effects of microencapsulation and microemulsion on antibacterial activity of natural antibacterial substances

Abstract: The natural antibacterial components are mainly the essential oils and plant metabolites of some plants.Natural antibacterial has the advantages of high safety, but its poor dispersivity, heat instability and volatilization have always been a major obstacle to its practical application. Microemulsion and microcapsule technology can greatly improve these shortcomings In this paper, microemulsion and microcapsules were prepared with two natural antibacterial components of lavender essential oil and salicylic acid. The inhibition rate of lavender essential oil, lavender essential oil microemulsion, salicylic acid and salicylic acid microcapsules on Staphylococcus aureus, dynamic bactericidal curve and dynamic cell membrane damage curve were studied. The results of antibacterial experiment showed that lavender essential oil, lavender essential oil microemulsion, salicylic acid and salicylic acid microcapsule had good antibacterial ability. The salicylic acid microcapsule had the most remarkable antibacterial ability, and the inhibition rate of Staphylococcus aureus reached 99.9999% when the concentration of salicylic acid was 0.125%. At the concentrations of 400ul/l and 800ul/l, lavender essential oil and microemulsion can also inhibit Staphylococcus aureus from 1 to 2 orders of magnitude. The results of the dynamic antibacterial test showed that 1min could reduce 4log in salicylic acid, while the exposure time to 30min could be reduced by 5log. Through dynamic cell membrane destruction experiment, we can conclude that salicylic acid microcapsule has sustained release effect. The results showed that the application of microemulsion and microcapsule technology to improve the poor application of natural antibacterial may be an effective way for the application of natural antibacterial in the field of practice.
