摘要:烟草品质主要受钾元素的影响。本实验分别在全素营养下与缺钾营养下,对烟草上中下叶位涂抹 2% 的硝酸钾溶液或者蒸馏水,运用电热消解和火焰光度法测定钾元素含量,探究烟草不同叶位间的钾离子转运情况。结果表明:缺钾营养下,涂抹 2% 的硝酸钾的烟草各叶位钾含量均有上升,说明施钾处理后,钾向各叶位均有转运,其中向上叶位转运情况明显,但各叶位叶片的生物量差异不明显。全素营养条件下,涂抹硝酸钾溶液与涂抹蒸馏水之间的差异不明显,生物量差异也不明显。另外,根据烟草TPK1基因的mRNA序列设计特异引物,进行实时荧光定量PCR,探索TPK1基因在叶面的表达情况。试验结果有待跟进。

Absorption and translation of potassium ion in different leaf positions of tobacco under the Potassium deficiency stress
Abstract: The quality of tobacco was mainly affected by potassium. This experience explore potassium ion transport on the top third leaf of tobacco by daubing 2% potassium nitrate solution or distilled water in different leaf positions under the Total nutrient and less potassium nutrient, using digestion to analysis Potassium content. The result showed that Potassium content was increased in each different leaf position under the less potassium nutrient. This shows that there is translation of potassium ion in difference leaf positions when daubing 2% potassium nitrate. It is obvious that the potassium in the up leaf position. but the biomass had the same conclusion in daubing distilled water and daubing 2% potassium nitrate. In the total nutrient, there is no difference of biomass and potassium in each treatments. Next, designing two pairs of amplification primers based the mRNA sequence of TPK1 in tobacco and establishing real-time fluorescent quantitative PCR, quantitative analysis was still followed up.
