蝉花醇提物抑菌活性及稳定性研究

摘   要:蝉花是中国传统虫生性药用真菌,有研究表明蝉花中的多糖、多球壳菌素和抗菌肽等都具有抑菌活性,并且拥有较高的抗氧化能力。本实验通过牛津杯法来测量蝉花醇提取物的抑菌圈大小,研究其抑菌图谱,从而分析其抑菌作用,并从酸碱性、温度和UV三个方面进一步探究抑菌活性物质受制因素。此外,分别通过DPPH清除实验,邻苯三酚自氧化法和Fenton法来测量蝉花醇提物及其有机萃取相的抗氧化能力,从而评判蝉花醇提取物的抗氧化能力。本实验结果表明蝉花的醇提取物对多数细菌均具有抑菌效果,但是针对真菌的影响较低。在蝉花醇提取物耐受性实验中,提取液对于酸碱性较为敏感,数据显示其能够耐高温和低温,对于紫外线不敏感。从本实验中可以看出蝉花醇提取物对DPPH自由基的清除率较高,对于醇提取液逐级分离萃取物也存在较高的抗氧化能力,另外数据显示蝉花醇提液对羟基自由基和超氧阴离子都具有一定的清除能力,是一种良好的抗氧化剂。

Abstract:Cordyceps cicadae is a traditional Chinese insecticidal medicinal fungus. Some studies have shown that polysaccharides, myriocin, and antimicrobial peptides in Cordyceps.c have antibacterial activity and have high antioxidant capacity. In this experiment, the Oxford cup method was used to measure the size of the inhibitory zone of the Cordyceps.c of ethanol extrac. and its antibacterial spectrum was studied to analyze its antibacterial activity. The antibacterial active substance was further explored from three aspects of restricted factors as pH, temperature and UV. In addition, DPPH scavenging assay, pyrogallol autoxidation method, and Fenton method were used to measure hydroxyl radicals to measure the antioxidant capacity of ethanol extracts and organic extracts, thereby evaluating the resistance of extracts of ellagitol. Oxidation capacity. The results of this experiment showed that the alcohol extracts of Cordyceps.c had antibacterial effects on most bacteria, but the effect on fungi was low. In the experiments on the tolerance of extracts of squid alcohol, the extracts are more sensitive to change of pH, and the data show that they can withstand high temperatures and low temperatures, and are insensitive to ultraviolet rays. From this experiment, it can be seen that the alcohol extracts of Cordyceps.c has a higher DPPH free radical scavenging rate, and there is also a higher antioxidant capacity for the stepwise separation of the extract from the alcoholic extract. The data also show that the alcohol extracts of Cordyceps.c has a hydroxyl group. Both free radicals and superoxide anions have certain scavenging capabilities and are good antioxidants.
