转pdmAB基因拟南芥修复土壤中取代脲类除草剂的研究

摘要:转基因植物修复技术是一项绿色环保的环境修复技术,将一些有机污染物降解功能基因导入植物中,获得转基因植株,增强植物在污染环境中的抗性及降解能力,达到去除污染物和修复环境的目的。取代脲类除草剂广泛使用造成的环境污染是一个全球问题,已引起了人们的极大关注。在本论文中,我们首次将来自于Sphingobium sp. strain YBL2取代脲类除草剂降解的关键基因pdmAB在拟南芥中表达。与对照相比,转pdmAB拟南芥表现出了更强的抗性。转基因拟南芥在含15mg/L异丙隆(IPU)的培养基和土壤中都能正常生长,而对照不能生长,最终死亡。此外,转pdmAB拟南芥在30天内能够吸收培养基和土壤中66%左右IPU,并降解成3-(4-异丙基苯基)-1-甲基脲(MDIPU)及少量的3-(4-异丙基苯基)脲 (DDIPU)。我们的研究表明转pdmAB植物通过表达修复IPU污染土壤能成为一种有效且环境友好型的方法。

Phytoremediation of Phenylurea Herbicides in Soil by Transgenic Arabidopsis thaliana Expressing pdmAB

Abstract:Phytoremediation of transgenic technology is green and environmentally friendly. Some genes have the ability to degrade contaminant were introduced into plants. It enhances the ability of resistance and degradation in a polluted enviroment so as to remove the contaminant and protect enviroment. The environmental pollution caused by widely used PHs has raised great attention. Through this research, we introduced key genes pdmAB that was from Sphingobium sp. strain YBL2 into Arabidopsis thaliana.. Compared to the control, Arabidopsis thaliana that was transferred pdmAB has showed more resistant to the adverse situation. Transgenic plants can grow normally in the culture medium or soil contained 15mg/L IPU, while the control cannot and dead finally. In addition, transgenic plants can absorb about 66% IPU in the medium or soil in 30 days and degrade it into MDIPU even a little DDIPU.
