摘要:为构建良好的西红花快繁体系,进行了西红花组培快繁技术研究,包括愈伤组织诱导、丛生芽诱导和球茎诱导各阶段的最适培养基。对染菌率、出芽率进行了观察记录。结果表明,将西红花球茎切块以及侧芽基部为外植体,以MS培养基为基本培养基,添加不同激素浓度及不同激素配比,于22-25 ℃黑暗培养诱导愈伤组织;将愈伤组织转入MS+6-BA3.0mg/L+NAA0.5 mg/L+白糖30g/L 培养基中, 于22-25 ℃暗培养诱导丛生芽;丛生芽切成单个不定芽转入MS+6-BA2.0mg/L+NAA0.5 mg/L+白糖30g/L+活性炭3g/L培养基, 于22-25 ℃见光培养诱导球茎。
Abstract:In order to construct a good safflower rapid propagation system, the safflower tissue culture propagation technology was studied, including the optimum medium for callus induction, cluster shoot induction and bulb induction. The observations were recorded on the rates of germination and budding.The results showed that the safflower bulbs and the lateral bud base were explants. The MS medium was used as the basic medium, and the concentrations and ratios of different hormones were added to induce the callus at 22-25 °C.;The callus was transferred into MS+6-BA3.0 mg/L+NAA 0.5 mg/L+sugar 30 g/L medium, and the clustered shoots were induced at 22-25 °C in dark culture;The shoots were cut into inpidual adventitious buds and transferred to MS+6-BA 2.0 mg/L+NAA 0.5 mg/L+sugar 30 g/L+activated charcoal 3 g/L medium, at 22-25 °C see light culture induced bulbs.
